Summary
We used microarray to create a normal cell landscape for the myeloid arm of the hematopoietic system.
Overall Design
Mononuclear bone marrow cells from heathly donors were enriched for 34+ and hematopoietic stem and progenitor cells were sorted by FACS. Bone marrow cells were aspirated from the posterior iliac crest of healthy subjects, according to the standard protocol of the Department of Hematology, Rigshospitalet University of Copenhagen (Denmark), and in accordance with the Declaration of Helsinki. All samples were obtained after informed consent according to guidelines established by the local ethics committee, which are in accordance with the Declaration of Helsinki. Hematopoietic stem cells (HSCs) and hematopoietic progenitors (HPCs) with the following immunophenotypes were purified essentially as described in earlier1-3 paper using a FACS Aria I cell sorter (BD Bioscience): HSCs: Linâ, CD34+, CD38â, CD90+, CD45RAâ, multipotent progenitors (MPPs): Linâ, CD34+, CD38â, CD90â, 45RAâ), common myeloid progenitors (CMPs; Linâ, CD34+, CD38+, CD45RAâ, CD123+), megakaryocyte-erythroid progenitors (MEPs; Linâ, CD34+, CD38+, CD45RAâ, CD123â) and granulocyte-macrophage progenitors (GMPs; Linâ, CD34+, CD38+, CD45RA+, CD123+).
